If you want to hike to the Sirena ranger station in Corcovado Park you have to go to the Puerto Jimenez area. You can stay in Puerto Jimenez, Dos Brazos de Rio Tigre, Cabo Matapalo or Carate. I would highly recommend 3 days, not 2. You need a guide, permits, reservations... A tour operator in Puerto Jimenez (for example Surcos Tours or Osa Wild) or your hotel can arrange the trek for you. It is not cheap. We paid $500 for 2 people (2015). You can choose between Los Patos-Sirena-Carate or Carate-Sirena-Carate. We did the latter. It is 20 km from Carate to Sirena, the second day you get up early (best time to see a tapir) and the whole day you will be hiking on the trails around Sirena, looking for wildlife, going to the river for a swim... Then the third day you hike back out. Carate to Sirena is partly on a jungle trail, partly on the beach. Los Patos to Sirena is a bit farther, harder and muddier and that part is all in the rainforest.
From Drake Bay you can't hike to Sirena but boat tours are offered. The boat brings you to Sirena, you hike some of the trails there and the boat takes you back. Most people do that as a day trip. You can also stay overnight but it is expensive.
