We will be arriving at Changi Airport at 6:10 in the morning. Will the ticket office be open then for us to be able to buy a tourist pass train ticket so that we can take the train to our hotel?

If you have just flown in from South Africa then you are doing yourself a favour to just grab a cab. Quite apart from having to wait until the station office opens, after intercontinental flights the taxi ride saves time and is a lot more comfortable than using the MRT on arrival.
I will use the MRT but only if I am light on luggage, in a hotel near a station, away from rush hour if using major nexus stations and only from a relatively local regional flight. From the UK, for example its taxis every time.
Are you sure the Tourist Pass is the best option for you over Ezlink or for short stays just buy a couple of tickets as you go.....the Tourist Pass is comparatively expensive for most users.
I just looked at the Ezlink option and that might help save cost if we take a cab from the airport.

Taxi in singapore is absolutely affordable.
You did not say when you are arriving. The EW Line (train line that links the airport to the city) is presently undergoing major maintenance work. There will be days when the entire line is closed and days where there will be shorter operational hours.
So as others have said, this is one more reason to take a taxi from the airport.
Yes luggage and the MRT,that time of day, could be a nightmare. Lots of people early in the mornings, LOTS. Plus who wants to hang around waiting for a ticket office to open ?
Plus when you get to your destination station, some of the exits are a major trex dragging heavy luggage, then you’ve got to get to your hotel.
Be kind to yourselves a cab will cost less than 30 bucks, worth every cent. imvho.
