I'm not aware of any single-day tour that does that route. As a generalisation, one-way tours are quite uncommon in Australia. However, it's the main highway between the state's two largest cities and the main bus companies do it twice a day:
Actually I will stay at Launceston for 4 days in Nov and another 5 days in Hobart. Given that it take around 2.5 hours from Launceston to Hobart and the highway bus fare is around A$35, I'm looking for some day trip at the mid-point, such as Wineglass Bay, Honeymoon Bay. Starting from Launceston and drop me at Hobart.

It is extremely complex and time-consuming to travel around Tasmania by public transport. However, you can get from Launceston to Bicheno and from Bicheno to Hobart with Calow's: http://www.calowscoaches.com/

However, your prospects of doing this complex journey in a single day are very slim.

Sorell Fruit Farm is open in November. http://www.sorellfruitfarm.com/
Several bus companies go through Sorell, however, you would have to walk the last three km to the farm. Ask directions from anyone in Sorell.
