the one day itinerary without a car on the upper right of your screen is a good starting point. also lots of previous posts on this topic--review a few for ideas and post back with more specific questions.
Which of the 302 activities would you recommend ?
I would not miss some time at PIke Place Market, the Space Needle, a ferry ride to Bainbridge Island and some time on the beautiful waterfront. The Ride the Ducks Tour is also a fun way to see Seattle. Try to get up to the Space Needle at night, the city lights are beautiful! If you would like some dining recs for the area, let me know, we have several favorites. We were just in Seattle in May and had a fantastic time. We were able to get over to Whidbey Island for a night at the Inn @ Langley which was very memorable.

Don't miss Pike Place Market and wataching the flying fish activity at the fish place at the far end of the market. They give quite a show.
Rachel the famous piggy bank is near there. Be nice and feed her - money.

You can walk down to see the first Starbucks Coffee Shop which is interesting. It's near Pike Place Market There was an excellent bakery near there which will go well with your coffee. There are several street vendors and activities in that area. A good restaurant we found in that area is Cutters and it is right on the water also with good views.
Our second day we went on the Duck tour leaving from near Macy's and had lots of fun with that. There's a free public bus that covers certain blocks downtown making it easy to get around.
We weren't prepared for the hilly streets in Seattle - similar to San Francisco.
